The name ‘gluten’ has been applied indiscriminately to the grain proteins of various grains, including maize, and difficulties of nomenclature have resulted. The prolamins of rye, triticale and barley (sometimes also oats) are sufficiently similar in sequence to wheat gluten that people with coeliac disease must avoid them. However, only wheat gluten has the appropriate rheological properties to sustain the production of good quality leavened bread. This property results from the balance of the component protein classes of gliadin (single polypeptides) and glutenin, which contains very large disulfide-linked aggregations of polypeptides, whose overall molecular size is a major determinant of dough strength (see Chapter 5).

Cereals are undoubtedly full of nutrition, but unfortunately, the refining process degrades their quality. The degree of milling, polishing, and refining to some extent decides the nutrient content of cereals. Some nutrients are lost during food preparation, especially vigorous washing, soaking, and cooking methods, which result in the depletion of the nutrients on the skin of the grains.The development of genomic and proteomic technologies has had a massive impact on our understanding of wheat grain proteins and will continue to do so in the future. The abundance of cereal grain proteins made them an attractive target for cloning cDNAs and genes, starting in the early 1980s (Bartels and Thompson 1983, Forde and Miflin 1983, Kasarda et al 1984). Many subsequent studies have provided full amino acid sequences of all of the major types of wheat gluten protein, allowing their structural and evolutionary relationships to be reassessed and placed in the wider context of plant protein families (Shewry et al 1986, 2002a; Jenkins et al 2005). Furthermore, the availability of massive libraries of expressed sequence tags (ESTs), corresponding to individual mRNAs, provides immense opportunities to the inquisitive researcher. The sequences of more than a million wheat ESTs are currently available (January 2007) in the EST database (dbEST, www.ncbi.nlm.nih.gov/dbEST/index.html), many derived from mRNAs expressed in the developing grain. They therefore provide an invaluable resource for gene discovery.Other types: Maize, rice, and wheat together make up 87% of total grain production all over the world.
